A review by four leptin researchers that draws the distinction between leptin resistance as a cause versus a consequence of obesity. The key argument, in animal models: in diet-induced obesity, cellular leptin resistance appears more a consequence of the obese state than its initiating cause, though ”It is debated to what extent”. The resistance then limits leptin’s control of adiposity, ”thereby magnifying the extent of weight gain”. The paper’s own methodological point is that the question of cause versus consequence ”remains largely unanswered”, because measurements taken in already-obese animals cannot separate them. The paper maps four specific molecular mechanisms (SOCS3, PTP1B, ER stress, inflammation) through which obesity attenuates leptin signaling, making the abstract concept of “leptin resistance” concrete and mechanistic.
